Brayan Villarreal's (0-1) pitch to Alex Rodriguez got away from catcher Alex Avila allowing Jeter to score.
Ivan Nova started for New York, surrendering 11 hits and six earned runs in 5 1/3 innings.
The Yankees got to Detroit ace Justin Verlander for four earned on seven hits over six innings.
A-Rod had three hits and two RBI'S on the night, including solo-home run off Verlander. Russell Martin touched up Verlander for a two-run shot in the fifth.
For the Tigers, Austin Jackson had four hits and two driven in, while Prince Fielder also drove in a pair on two hits.
Mariano Rivera (1-1) worked a scoreless top of the ninth to earn the win, capping 3 2/3 innings of shutout ball for the Yanks' bullpen.
